针刺对海洛因成瘾戒断大鼠下丘脑-垂体-肾上腺系统的影响
Effect of acupuncture on hypothalamic-pituitary-adrenalsystem in heroin withdrawal rats

Abstract
Aim To assess the effect of acupuncture on anxiety-related behaviors after heroin-withdrawal, and explore its possible mechanism. Methods Heroin was injected subcutaneously according to the principle of daily increasing dose in rats of experimented group. The abstinence signs were precipitated by naloxone after drugs were stopped, and the animal model of heroin addiction was established. Acupuncture was continuously given for 7 days. The elevated plus maze test was used to observe anxiety level after heroin-withdrawal and the anti-anxiety effect of acupuncture. The concentration of corticosterone ( CORT ), adrenocortico-tropic hormone ( ACTH ) and arginine vasopressin ( AVP ) in the plasma of the rats were detected by radio-immunity. Expressions of Fos protein in the para-ventricular nucleus of rat hypothalamus were detected by immunohistochemistry method. Results Acupuncture significantly increased the percentage of entries in-to open arms and time spent on open arms( P <0. 01 or P < 0. 05 ). The radioimmunoassay results showed that acupuncture decreased plasma CORT, ACTH and AVP in heroin-withdrawal rats( P < 0. 01 or P < 0. 05 ). The immunohistochemistry results showed that the expression of Fos protein in hypothalamic paraventricular nucleus was decreased after acupuncture ( P < 0. 01 or P < 0. 05 ). Conclusions Acupuncture reduces anxiety level of heroin-withdrawal rats and modulates the hypothalamic-pituitary-adrenal system activities. These findings suggest that acupuncture at "shenmen" point may be useful as a therapeutic treatment in heroin withdrawal induced emotional disorders, and its mechanism may be related to modulating the activities of hypothalamic-pituitary-adrenal system.关键词
