Mechanism of DNA methylation in the regulation of heat shock-induced transcriptional inhibition of cyclin D1 expression.
摘要
Abstract
Objective To investigate the role of DNA methylation in heat shock -induced inhibition of human cyclin D1 expression. Methods The DNA methylation status and mutatan sites were detected by Methylmion Specific PCR ( MSP) and Stratagene kit, respectively. The effects of the methylation sites were evaluated by reporter gene analysis. Results Suggest that the + 65/67 sites of human cyclin Dl promoter were methylated under heat shock treatment. The heat shock response evaluated by reporter gene analysis was inverted when the putative methylation sites was mutated. Conclusion The heat shock - induced inhibition of cyclin Dl transcription involves the methylation of + 65/67 CpG sites.关键词
