摘要

Abstract

[Objective]A method for determining total saponin content in aqueous extract of tomatoes was developed to facilitate studies on tomato saponins. [Method]After zymohydrolysis, tomato slurry obtained by smashing was subjected to D-101 macroporous resin to get crude saponins. Esculeoside A was used as reference and vanillin-perchloric acid was applied as chromogenic reagent to determine total saponin content at 550 nm. [ Result ]The standard curve of esculeoside A showed good linearity in the range of 0.052-0.312 mg with R2=0.9958, and the average recovery of added sample was 99.53% with 0.52%(n=6) RSD. Total saponin content obtained in five batches of crude saponin extraction was found to be 46.75±2.09%. [Conclusion]The method developed in the present study was rapid, accurate and easy to be used,therefore would be good for determination of total sapnion content in water extract of tomatoes.
