摘要

Abstract

Delta l-pyrroline-5-carboxylate synthetase( P5CS) plays a critical role in improving the stress tolerance of plants. In this study, the full length cDNA sequence of P5CS gene was cloned from Alkaligrass ( Puccinellia chinampoensis) leaves using RT-PCR method, the primers were designed according to the homologous P5CS gene sequences of other plant species. Sequence analysis showed that the nucleotide sequence of this gene is 2 158 bp,containing a complete open reading frame and encoding 716 amino acids,Genebank ;HQ637435. Nucleotide and amino acid sequence analysis revealed that PuP5CS shares high identity with the orthologs from Triticum aestivum. And its signal peptide, hydrophobicity/hydrophilic, trans-membrane domain, secondary structure and main functional domains were predicted. Semi-quantitative RT-PCR analysis showed that PuP5CS expressed in different tissues, but the expression in root was lower,and in leaf much higher. Various elevated levels of PuP5CS expression have been detected when exposed to 4 different stress experimental treatment, and the results was not the same.
