Pingyangmycin-regulated expression of E-selectin in human venous malformation endothelial cells.

Abstract
Objective:To investigate Pingyangmycin (PYM) -regulated expression of E-selectin in human venous malformation endothelial cells (HVMECs) and mechanism of sclerotherapy of venous malformations with PYM. Method: Expression of the adhesion molecule E-selectin was studied in PYM -stimulated HVMECs by the means of cell ELISA and reverse transcription and the polymerase chain reaction (RT-PCR). Result: E-selectin was induced in a time-dependent fashion after stimulation with PYM. The expression of E-selectin on cell membrane was observed after 2 h exposure to PYM. The highest expression of E-selectin was observed at 6 h. E-selectin started to disappear at 8 h. No significant expression of E-selectin was detected after 24 h exposure. RT-PCR revealed the expression of mRNA of E-selectin started at 2 h. Maximal synthetic activity was observed at 6~8 h for E-selectin. Thereafter, synthesis slowly decreased. E-selectin was induced in a concentration-dependent fashion after stimulation with PYM. After exposure to 1 (μg / mL PYM, HVMECs showed maximal expression of E-selectin compared to other concentration of PYM. Synthesis activity was greatly restrained at 10 μg / mL or higher concentration. Conclusion: Expression of E-selectin in HVMECs can be induced by lower concentration of PYM in a time- and concentration- dependent fashion, and restrained by high concentration of PYM.关键词
