Abstract
Objective: To investigate the effect of arecoline on the expression and distribution of Ang II and one of it's receptors-AT1R in Hacat cells and oral mucosal fibroblasts (OMFB), and to further gain insight into the pathogenesis of OSF. Method: The specific SABC immunocytochemistry was used for detecting the expression of Ang II and AT1R rabbit anti-hu-man polyclonal antibodies in Hacat cells and OMFB after 48-hour exposure to arecoline of various concertrations. Result: ① Positive expression of Ang Ⅱ and AT1R was detected in normal Hacat of the four groups, and the exposure of Hacat to Arecoline at concertrations of 80μg/ml elevated Ang II and AT1R expression, the difference was statistically significant (P < 0.01). ②Negative expression of Ang II and AT1R was detected in normal Hacat and the cells exposed to arecoline of 20 μg / mL. Slightly positive expression was detected in the cells exposed to arecoline of 40 μg / mL. Positive expression was detected in Hacat exposed to areciline of 80 ug / mL. The difference was statistically significant (P <0.01). Conclusion: Arecoline at a certain concertration can upregulate Ang II and AT1R expression in Hacat, and the expression of these two protein was detected frequently in the cells which have morphological changes. These indicate that Ang II and AT1R may contribute to the pathogenesis of OSF.关键词
