摘要

Abstract

Objective To determine express of Kvl. 3,KCa3. 1 mRNA in lymphocyte derived from sponsta-neously hypertensive rat (SHR) and identify the relationship between lymphocyte Kvl. 3,KCa3. Land hypertension. Methods Male SHRs and normotensive Wistar rats were obtained from Experimental Animal Company of Beijing Vital River and Experimental Animal Center of Xinjiang Medical University. Peripheral blood were collected from SHRs and normotensive Wistar rats. Peripheral lymphocytes were separated from heparinized whole blood by standard Ficoll-Hypaque density gradient centrifugation. Real-time PCR was used to detect the expression of Kvl. 3,KCa3. 1 mRNA in lymphocytes. Results ( 1) The expression levels of Kv 1.3 mRNA in lymphocytes from SHRs were significantly increased compared with Wistar rats (P<0.05). (2)Compared with the controls, Kca 3. 1 mRNA were incresed in in lymphocytes from SHRs (P<0.05). (3)Linear correlation existed between systolic blood pressure and expression of Kvl. 3,Kca3.1 mRNA in lymphocytes from SHRs ( r - 0. 830,P = 0.003;r = 0.674,P = 0.03,respectively). Conclusion The lymphocyte Kvl. 3 ,KCa3. 1 mRNA expression are upregulated in SHR,which suggests that Kvl. 3 and Kca 3. 1 maybe contribute to the development of hypertension.
