Abstract

Ferredoxin(Fd)cDNA was amplified by RT-PCR from sugarcane leaves, and then it was ligated into pMD18-T vector and the corresponding sequences were analyzed. The results indicated that the fragment length was 688 bp, including Fd cDNA ORF encoding 151 amino acids. Amino acid sequence analysis showed that there were some acidic amino acid residues, and those amino acid sequences between 76 to 101 were highly conserved. Sequences alignment showed that the homology of cDNA sequence between sugarcane and maize Fd gene was 93.68%, while the homology of amino acid between them was 95.39%. Relative protein molecular weight was 16.05 ku, PI was 4.72, prohydrophobicity from-2.567 to 1.344. The molecular structure prediction results suggested that the protein contained 13.9% a-helix, 25.8% p-meander and 60.3% loop. The sequence cloned in this study was named as Fdl, which has been submitted to GenBank with Accession number of GU320210.1. The present study should lay a base for the function study of this gene in sugarcane.
