Abstract

By using amplified fragment length polymorphism (AFLP) and specific-sequence amplified polymorphism (SSAP) markers, this paper studied the genetic diversity and genetic structure of four Hordeum brevisubulatum (Trin. ) Link populations mainly comprised of salt-resistant mutants from the Songnen Plain of Northeast China. Due to the slight difference of SSAP and AFLP markers in their principles, there existed small differentiation in the analytical results, but the results had a high level correlation by Mantel test ( r = 0. 88 , P<0. 01). The AFLP and SSAP analyses showed that the total proportion of polymorphic loci (P) was 23. 3% and 37. 7% , Shannon index was 0. 10 and 0. 14, Nei' s gene diversity index was 0. 07 and 0. 09, and gene differentiation coefficient ( Gst) was 0. 49 and 0. 44, respectively. These results indicated that there was a high level genetic diversity within the four populations, though they all formed separately from a mutant line. The AMOVA and cluster analyses also indicated that the population genetic differentiation mainly existed within populations, though there was a high level genetic difference among the populations.
