Abstract

The soil samples collected from a saline-alkali land in Daqing City of Heilongjiang Province, Northeast China were properly diluted, and smeared on a high-alkali culture medium. Forty one bacterial strains were isolated and purified. Further screening of the strains by progressively increasing the salt and alkali concentrations in the culture medium respectively or simultaneously resulted in the discovery of a saline-alkali tolerant strain which could even grow in a medium with pH 13 and containing 200 g ? L"1 of NaCl. The strain belonged to both extreme halo-phile and extreme alkaliphile. The study of the biological characteristics and salt-alkali removal efficiency of the strain under different culture conditions including carbon source, nitrogen source, carbon/nitrogen ( C/N) ratio, initial pH value, and temperature indicated that for the growth and the salt-alkali removal efficiency of the strain, beef extract was the best carbon source, peptone was the best nitrogen source, and the optimum C/N ratio was 4:1. For the growth of the strain, the optimum initial pH value was 9. 5 , and the optimum temperature was 20 °C ; for the removal of salt and alkali by the strain, the optimum temperature was 30-35 t.
