Abstract
Objective To study the time-dependent expression of S100β following acute mechanical injury of brain stem in rabbits. Methods The brain stem of 36 health rabbits injured are dissected after injury 0,2,4,8, 16,24,48,72 h. Normal rats as control. The mRNA expression of S100β was detected by real-time PCR and the expression of S100β protein was detected by immunohistochemistry. Results S100β mRNA expression induced within 24 h after injuried, S100β mRNA expression increased at 24 h time point and subsequently reduced. S100β protein uniformly distribute in cell body and apophysis of astroglial cells in brain stem of rabbits after injury 0 h. Two hours after mechanical injury of brain stem, astroglial cells are destroyed with apophysis disappearing, then with time pass by the concentration of S 100β increasing, at 24 h after injury the concentration reaches the peak. Then the concentration decreased but is higher than that at 0 h (P < 0.05). Conclusions S100β mainly lies in neuroglial cell, is sensitive biochemical index and is golded index of early hypoxic-ischemic brain damage and prognosis. Otherwise S100β also apply for concluding injury age of forensic injury of central nervous system, so offer experiment basis for the forensic identification.关键词
