SV40 large T antigen induces human-derived lymphocyte malignant transformation

Abstract
Objective To study the effects of SV40 large T on human-derived lymphocyte transformation. Methods Peripheral blood mononucleate lymphocytes were conducted with the retroviral vector plasmid pBabe-SV40 large T. The cells morphological changes were watched by inverted microscope. The protein of large T antigen was detected by Western blot. Lymphocyte surface molecules were identified by flow cytometry. The cell karyotype was analyzed by G-banding. The cell proliferation was described by growth curve. The soft agar assay was performed to examine the cells malignant transformation. Results Some significant morphological changes were observed in the SV40LT gene transfected human-derived lymphocytes is accompanied by transition from suspension to adherent growth. The expression of large T antigen was found in the transfected cells. Moreover, 67% cells showed CD56 marker and were able to grow in soft agar. Conclusion SV40 large T antigen successfully induced a stable passaging and malignant transformation of lymphocyte with expression of CD56+.关键词
