The expressions of ARK5, MMP-2, and MMP-9 in gastric cancer and their relationships with the metastasis of gastric cancer

Abstract
Objective To investigate the expressions of ARK5 , MMP-2, and MMP-9 in gastric cancer and their relationships with the invasion and metastasis of gastric cancer. Methods The expressions of ARK5 , MMP-2, and MMP-9 in 66 cases of gastric cancer tissues and 20 cases of normal gastric tissues were measured by immunocytochemistry; ARK5-siRNA was transfected into SGC-7901 cell lines by liposome mediated-method (siARK5/SGC7901 group), with SGC-7901 cell lines transfected by SCRsiRNA as control group. The expressions of ARK5 , MMP-2, and MMP-9 were measured by Western blotting analysis. The in vitro invasion ability was examined by Transwell invasion assay. Results The expression rates of ARK5 , MMP-2, and MMP-9 in gastric cancer tissues were 68% , 76% , and 68% , respectively, and there were significant differences among them (P<0.05). The expression of ARK5 was positively correlated with the depth of invasion , the degree of cellular differentiation, theTNM staging, and the lymph node metastasis (P<0.05), but was not related to the age, sex, and tumor diameter (P>0.05). The protein levels of ARK5 , MMP-2, and MMP-9 in siARK5/SGC7901 group were obviously decreased and the in vitro invasion ability were significant decreased (P<0.05). Conclusions There were significant correlations among the expressions of ARK5, MMP-2, and MMP-9. The expression of ARK was closely correlated with the invasion and metastasis of gastric cancer, wich can be used as a prognosis index and chemical treatment targets for gastric cancer.关键词
