Abstract
BACKGROUND: Cancer stem cells marker expression in different environments differ greatly because the microenvironment can impact the expression of proteins.OBJECTIVE: To discuss whether different cultures in vitro have influence on the expressions of cancer stem cells marker CD44, ESA and CXCR4.METHODS: Tongue cancer stem cells TCA8113 cell line was obtained. In vitro, three different microenvironments were established: stem cell culture medium, conventional standard medium and conventional standard medium supplemented with adriamycin. The expressions of three proteins in TCA8113 tongue carcinoma cells were detected by immunohistochemistry (IHC) and flow cytometry (FCM).RESULTS AND CONCLUSION: In all microenvironments, IHC showed the expressions of CD44 and ESA were"++". In stem cell culture medium and the other microenvironments, the expressions of CXCR4 were "±" and "+" respectively. FCM found the expressions of CD44 and ESA were above 90% in all microenvironments; compared with conventional standard medium, the expression of CXCR4 was super low in stem cell culture medium, and recovery when tongue cancer cells reversed into conventional standard medium from stem cell culture medium; the expression of CXCR4 was increased in conventional standard medium supplemented with adriamycin. The expressions of cancer stem cell markers were various in different mircoenvironments; it needs to take various markers into account and combine with the experessions of markers on cancer cells in vivo to identify cancer stem cell markers. Different kinds of cancer stem cells may accumulate together by different microenvironments model.关键词
