microRNA-196a2单核苷酸多态性对靶基因IL-2表达的影响
Influence of microRNA - 196a2 single nucleotide polymorphism on expression of target gene interleukin - 2

Abstract
Objective To observe the influence of single nucleotide polymorphism( SNP )on the expression level of mature microRNA( miRNA )- 196a2 in human keratinocyte HaCaT cell and on the binding capacity of predicted target messenger RNA ( mRNA )interleukin - 2( IL - 2 )3 untranslated region( UTR),and to discuss whether miRNA - 196a2 SNP has any correlation with autoimmune diseases. Methods Transfection and construction of miRNA - 196a2 - T/C( wild type/mutant type Expression plasmids was made in HaCaT cells. The expression effects were detected by RT - PCR. Co - transfection was made between the two gene types of expression plasmids and the report gene plasmid of predicted target mRNA IL - 2 3'UTR. Then the binding capacity was detected by the dual - luciferase report gene experiment,and the results were statistically analyzed by SPSS 17. 0 software. Results MiR - 196a2 - T/C expression plasmids and IL - 2 3UTR report gene plasmids were successfully constructed. In the HaCaT cells,there was no significant difference in the expression level between the wild and mutant type of miRNA - 196a2 expression plasmids( P > 0. 05 ). IL - 2 was the target gene of miRNA - 196a2. The binding of miRNA - 196a2 and IL - 2 3UTR was influenced by SNP( P < 0. 05 ). Conclusion MiR - 196a2 bound with IL - 2 can adjust its post - transcriptional. Thereby it participates the immunological reaction and may influencethe proceses of autoimmune diseases.关键词
