摘要

Abstract

Objective To observe the expressions of aquaporin4 (AQP4) and vascular endothelial growth factor(VEGF) mR-NA in human retinal Miiller cells cultured under hypoxia in vitro. Methods The human Muller cells were cultured using the enzymatic digestion, identified by transmission electron microscope and immunofluorescence staining. The expressions of AQP4 mRNA and VEGF mRNA in human retinal Muller cells under hypoxia induced with CoCl2 were detected by RT-PCR. Results The positive expressions of glutamine synthetase, neuroglial fiber acidic protein, ot-smooth muscle actin, intermediate filament vimentin were showed in over 95% cells under fluorescene microscope. The expressions of AQP4 mRNA and VEGF mRNA in Muller cells were both increased with a partial dose and time dependent manner,especially of VEGF. The mRNA expression of VEGF was reached peak at 12 hours and AQP4 was at 24 hours. Conclusion Both AQP4 and VEGF may have close relationship with cell edema under hypoxia and the effect of VEGF may be earlier and stronger,which demonstrated that they may play roles in edematization of hypoxic retinopathy.
