摘要

Abstract

Objective:To invesligale the polential of Ku80 small-interfering RNA (siRNA) Lo enhance cisplalin-induced apop-Losis. Methods:Ku80 mRNA and prolein expression levels were investigaled by reverse transcription-PCR and Western blot analysis,re-spectively. Ku80-specific siRNA was synthesized and Lransfected inlo cisplatin-resistant human lung adenocarcinoma cell line A549/ DDP prior Lo LreaLment with cisplalin. Cell survival was assessed by MTT assay. Apoptosis was assessed using flow cylomelry analysis. Caspase-3 activity was detected by speclropholomeler. Results:The Ku80 mRNA and prolein expressions were significantly elevaled in A549/DDP cells compared wilh the parenlal A549 cells. The Ku80 mRNA and prolein expressions were significantly downregulaled by si-Ku80 in A549/DDP cells. Treated wilh cisplalin for 24 h, the inhibilion rales of si-Ku80 cells decreased significantly compared wilh in conlrol group and si-Scramble group. After 24 h trealmenl wilh 6 μg/ml cisplalin, apoplosis rale in si-Ku80 group was increased compared wilh the conlrol group and si-Scramble group ( P<0. 05 ) ; aclivilies of Caspase-3 significantly increased in siKu80-A549/ DDP cells as compared wilh conlrol cells and si-Scramble cells (P<0. 01). Conclusion:Ku80 may correlate wilh the chemoresislance lo cisplalin in lung cancer. Ku80-specific siRNA enhanced cisplalin-induced apoplosis in the cisplalin-resislanl A549/DDP cells.
