德都红花-7味散对CCl4诱导实验性肝纤维化疗效和IL-1、IL-6、TNF-α的影响

Abstract
Objective:To observe the Deduhonghua-7 pill's affeclion on the degree of CC14 induced experimental hepatic fibrosis and its effect on cytokines IL-1, IL-6, TNF-α, and Lo explore its mechanisms. Methods:The rats were randomly divided into 5 groups,except the normal conlrol group, other groups were injected intraperiloneally with 30% (v/v) solution of CC14 in olive oil Lo cause hepalic fibrosis;modelling the same lime, the rals in Deduhonghua-7 pill high dose group,low dose group and colchicine posilive conlrol group were given inlragaslric adminislralion Deduhonghua-7 pill wilh 0. 62 g/kg and 1. 04 g/kg and colchicine 0. 4 mg/kg respectively each day. After the seven weeks,collected the serum and liver lissue,lhen perform the enzyme-linked immunosorbenl assay (ELISA) lo deled the liver fibrosis indexes and serum cylokines TNF-α,IL-l ,IL-6 levels and perform HE,Gomori,Masson slaining lo observe pathological changes of liver tissue. Use the " Consensus of the diagnosis and trealmenl of liver fibrosis" and " Semi-quantitative assessment slaging system Ishak of liver fibrosis" established at Xi'an council 2000, lo grading, staging and scoring the liver fibrosis. Results:Compared low dose group wilh model group, the serum and the conlent of liver lissue HA,LN,PCⅢ , IVC, Hyp,inflammatory activity and fibrosis semi-quanlilalive, the contenl of serum cylokines TNF-α,IL-1, IL-6 reduced significantly( P<0. 05 ) ; the contenl of high dose group's serum HA and IVC, liver tissue LN and IV C, inflammatory activity and fibrosis semi-quanlilalive, serum cylokine TNF-α reduced significantly (P<0. 05) ;conlent of posilive control group's liver lissue HA increased, IV C decreased, had significant difference( P<0. 05 ) , inflammatory activity and fibrosis semi-quanlilalive showed no significant difference( P>0. 05 ) , contenl of serum cylokines TNF-α,IL-1,IL-6 increased,had significant difference(P <0. 05). Compared wilh normal conlrol group,serum HA,PC Ⅲ , Ⅳ C, liver tissue LN, PC Ⅲ , IV C in Deduhonghua-7 pill low dose group and serum PCⅢ and IV C, liver lissue IV C in Deduhonghua-7 pill high dose group showed no significant difference ( P>0. 05 ). Conclusion: Deduhonghua-7 pill can significantly improve CC14 in-duced experimental liver fibrosis pathology,reduce hepalic fibrosis indexes HA,LN,PCⅢ , IVC and cylokines IL-1 , IL-6,TNF-α in serum, and have and liver fibrosis effect.关键词
