淫羊藿次苷Ⅱ对小鼠脾淋巴细胞体外增殖的影响及其机制研究
Effects and mechanism of IcarisideⅡ on the proliferation of mice splenic lymphocytes in vitro

Abstract
Objective:To study the effects of Icariside Ⅱ ( ICS Ⅱ ) on the proliferation of mice splenic lymphocyles in vitro and elucidale mechanism of its immunoregulation effecls. Methods:Mice splenic lymphocyles were deall with differenl concenlralion of ICS II and concanavalin A. The effecls of splenocyle proliferalion was delecled by CCK-8 melhod in order to selecl the most effeclive concenlralion of ICS Ⅱ. After the selection of concenlralion, cells were deall wilh it at differenl lime poinls. The effecl of cell proliferalion was delecled by CCK-8 melhod . Cell cycle distribution was observed by flow cylomelry (FCM) also al differenl lime poinls. The changes of inlracellular calcium concenlralion of cell were measured by fluorescence microplale reader. Results:Afler cultivation of cells for 48 hours with 9 concenlralion gradients of ICS II , a range of 10 -10 mol/L ICS II could promote the proliferation of cells. This promolion presented concenlration-dependenl effects . A range of 10 -7-10 -4 mol/L ICS II presented the inhibitory effects. ICS Ⅱ of 10-9 mol/L was found lo promote the proliferation of cells al different lime points and this effect was belter than those wilh ConA only. FCM tests could reveal the fact that ICS Ⅱ can promote cells lo enter into the milosis by reducing the percentage of cells which in GO/ Gl phases while rising the percentage of cells in S and G2/M phases, with the effect being mosl marked in 24 h. The results of the tesls taken by the fluorescence microplale reader can reveal that the fluorescence value of the ICS Ⅱ group was increasing and the effect was mosl marked in 12 h. Conclusion:Different concentrations of ICS Ⅱ could promot the proliferation of cells, presenling the concenlration-dependenl and time-dependenl effect. The mechanism of this effect might through increasing the calcium concenlralion in cell plasm and promoling cells cycle to enter into the S and G2/M phases from G0/G1 phases.关键词
