摘要

Abstract

Objective:To study the effect of human trophoblasts on the production of IL-10 and TGF-β in decidual γδT cells via the CXCL16/CXCR6 pathway. Methods:10 samples of villus and deciduals were collected from normal early pregnant women, then we isolated the trophoblasts and decidual γδT cells respectively. We detected the concentration of CXCL16 in the supernatant of trophoblasts by ELISA and the CXCR6 mRNA of decidual γδT cells by RT-PCR. We established the co-culture system of trophoblasts and decidual γδT cells, then detected the expression of intracellular cytokines IL-10 and TGF-β in the γδT cells by FCM. Results:The trophoblasts from early pregnant women could continually secrete CXCL16 in vitro, with a time-dependent effect, and decidual γδT cells could express CXCR6. In the direct co-culture condition, the trophoblasts significantly promoted the production of IL-10 and TGFβ in decidual γδT cells. This stimulatory effect of trophoblasts was partially abolished by CXCL16 neutralizing antibody. However, when decidual γδT cells were indirectly cocultured with trophoblasts, the production of IL-10 and TGF-β in decidual γδT cells was similar to the control. Conclusion:Human trophoblasts might promote IL-10 and TGF-β production in decidual γδT cells via CXCL16/ CXCR6 pathway, which is beneficial to the Th2 bias phenomenon at maternal-fetal interface and maintaining maternal immunotoleranee toward the fetus in early human pregnancy.
