UV法与HPLC法测定马尾松松针中莽草酸含量的比较
Content Determination of Shikimic Acid in Leaves of Pinus massoniana by HPLC and UV Method
摘要
Abstract
[ Objective]The determination results of shikimic acid in leaves of Pinus massoruana by HPLC and UV were compared. [ Methods] The separation was performed on Alltima NH2(150.0 mm×4.6 mm,5.0 μm).The mobile phase consisted of Acetonitrile-2%H,PO,90: 10) at the flow velocity of 1 ml/min. The test wavelength was 213 nm and the temperature of column was 25 ℃. The test wavelength was at 213 nm after sample solution had been diluted with 0.2% H3PO4. [ Results ] The linear rang of ghikimic acid was 0.29 - 2.88 jig/ml (R2 = 0.999 9) by HPLC method. The average recovery rate was 98.30% (RSD =0.74% ,n =5). The linear rang of shikimic acid was 4.7-23.0 μg/ml (R2 =0.999 8) by UV method. The average recovery rate was 97.25% ( RSD = 1.07% , n = 5). [ Conclusion ] The precision, repeatability, stability and recovery rate of UV and HPLC were satisfied. These two methods could both-use for the determination of the content of shikimic acid in leaves of Pinus massoruana.关键词
