Effect of immunomodulating peptide on ovarian cancer skov3 cell apoptosis and related gene expression
摘要
Abstract
Objective To study the effect of immunomodulatory peptide ( PGPIPN ) on human ovarian cancer skov3 cell apoptosis and related gene expression. Methods That skov3 cells cultured in vitro were incubated with different concentrations of PGPIPN ( 10 -6 ~ 10 -2 g/L ) for 48 h. The cDNA microarray technology was used to screen apoptosis related gene. Bax, Bcl-xl, Caspase-3 mRNA expression levels were detected by real-time fluorescent quantitative polymerase chain rection ( PCR ) method. Bax, Bcl-xl, Caspase-3 protein expression levels were detected by Western blot. Results Compared with the control group, the expressions of Bax and Caspase-3 were increased with the increase of the concentration of PGPIPN ( P < 0. 05 ). The expression of Bcl-xl was decreased with the increase of the concentration of PGPIPN ( P < 0. 05 ). Conclusion PGPIPN can induce the cell apoptosis by increasing the expression of Bax and decreasing the expression of Bcl-xl, this process may be related with Caspase-activated.关键词
