Abstract
Objective To study the expression and significance of peripheral blood Th17 cells in patients with sta-ble chronic obstruetive pulmonary disease( COPD ) and normal lung function smokers. Methods Choose non-smokers with normal lung function ( A group, 18 eases ), smokers with normal lung function ( B gmup, 20 eases ), sta-ble mild to moderate ( C gmup, 19 eases ) and severe ( D group, 15 eases ) COPD patients in our study. Percent-age of Th17 eells in peripheral blood mononuelear eells ( PBMC ) was determined by flow cytometry. IL-17 levels in plasma and PBMC supernatant after stimulation were measured by EL1SA. Results The pereentage of Th17 cells, the levels of 1L-17 in peripheral blood and PBMC supernatant after stimulation of group B, group C and group D were higher than those of group A( P <0. 01 ). The pereentage of Th17 cells and levels of 1L-17 in peripheral blood in group C and D were higher than those of group B( P <0. 01 ). The levels of IL-17 in plasma and PBMC supernatant after stimulation in group D were increased significantly compared with those of gmup C( P <0. 01 ). The level of IL-17 in PBMC supernatant after stimulation in group D was higher than that in group B( P <(). 01 ). In group C and D, the levels of IL-17 in plasma and PBMC supernatant after stimulation were negatively correlated with FEV1 ( % of predieted value ). The levels of IL-17 in plasma and PBMC supernatant after stimulation were positively cor-related with the pereentage of Th17 in PBMC. The pereentage of Th17 in PBMC was positively correlated with the years of smoking in all smokers. The pereentage of Th17 in PBMC had no relation with the yeal's of smoking in gmup C and gmup D. Conclusion The levels of Th17 cells and IL-17 in peripheral blood increased in patients with stable COPD and smokers with normal lung function. The pereentage of Th17 in PBMC associated with the years of smoking in smokers with normal lung function. In patients with stable COPD, the level of IL-17 in peripheral blood was negatively correlated with FEV1( % of predicted value ),the pereentage of Th17 in PBMC had no relation with FEV1( % of predicted value ) and the years of smoking.关键词
