Abstract
Objective: To investigate the effects of adenosine A2a receptor antagonist SCH58261 on generalised convulsive seizures induced by pentylenetetrazol and pentylenetetrazol-induced kindling. Methods: Generalised convulsive seizures and kindling models were induced in rats by intraperitoneal injection of pentylenetetrazol. Adenosine A2A receptor antagonist SCH58261 was given to rats and the treatment effect of SCH58261 was studied on the two epilepsy models. Results: Long-term administration of SCH58261 increased the latency of pentylenetetrazol induced clonic seizures but not inhibited the tonic seizures. Long-term administration of SCH58261 prevented the process of kindling and decreased severity of seizures on pentylenetet-razol-induced kindling model. Conclusion; Adenosine A2A receptor antagonist SCH58261 inhibits generalised convulsive seizures induced by pentylenetetrazol and pentylenetetrazol-induced kindling on rats.关键词
