培门冬酶抗人套细胞淋巴瘤细胞株Jeko-1细胞作用机制的研究
THE MECHANISMS OF ACTION OF ONCASPAR AGAINST JEKO-1 MCL CELL LINE
摘要
Abstract
Objective:To investigate the effects of oncaspar on proliferation and its possible mechanisms of human Jeko-1 MCL cell line in vitro. Methods: MTT assay was used to evaluate the inhibition of different concentrations of oncaspar treatment of Jeko-1 cells for 24 h and 48 h. Apoptosis and cycle distribution of Jeko-1 cells were detected by flow cytometry (FCM). SYBR Green I of real time fluorescent quantitative (qRT-PCR) to detect the change of Jeko-1 cells cyclinDl mRNA expression. Results: Oncaspar obviously inhibited the proliferation of the Jeko-1 cells, the inhibitory effect of 48 h group was significantly higher than that of 24 h group ( P <0. 05). FCM showed that the early apoptosis rate of treatment group was significantly increasing and the cells were blocked at G0/G1 phase ( P <0. 05) compared with the control group. qRT-PCR showed that oncaspar could significantly decrease expression of Jeko-1 cells' cyclin D1 mRNA , 2-ΔΔCT = 0. 42 + 0. 11. Conclusion: Oncaspar can inhibit Jeko-1 cells growth in vitro by inducing apoptosis and arresting cell cycle at G0/G1 phase, its mechanism maybe through decreasing Cyclin D1 mRNA expression.关键词
