Effect of Ulinastatin on the Lipopolysaccharide-induced Production of Nitric Oxide and mRNA Expression of iNOS in Macrophages

Abstract
Objective To investigate the effect of ulinastatin on the lipopolysacchaiide( LPS )-induced production of nitric oxide( NO ) and the mRNA expression of inducible nitric oxide synthase( iNOS ) in macrophages. Methods The RAW264.7 macrophage cells activated with LPS were pretreated with ulinastatin of different concentrations ( 100 - 10 000 U/ml ). The production of NO was determined by the Criess test. The mRNA expression of iNOS was determined by Real-time RT-PCR. Results Ulinastatin,especially in high concentration ranges( 1 000 -10 000 U/ml), inhibited LPS-induced production of NO( P < 0.05 ), decreased the mRNAs expression of iNOS( P < 0.05 ); There was no significant difference in NO and the mRNA expression of iNOS in LPS-induced RAW 264.7 cells between ulinastatin of low concentration 100 U/ml) and LPS alone( P > 0.05 ). Conclusion High concentration ulinastatin can inhibit the production of NO and mRNA expression of iNOS in LPS-induced macrophages, and the effect is correlated with the concentration of ulinastatin.关键词
