Abstract
Objective:To verify the antibacterial activities of the Saxifrage and provide the basis for its ingredient prescription at the same time through the in vitro antimicrobial effect of the three components of pyrogallol, hydroquinone and gallic acid methyl ester of Saxifrage. Methods: Constant dilution method was applied for the determination of the in vitro inhibitory effect of pyrogallol, hydroquinone and gallic acid methyl gallate on the five major pathogens leading to cow mastitis and orthogonal experiment was conducted to optimize its best ratio of the three components and obtain its validation. Results:The minimal inhibitory concentrations( MIC) of pyrogallol on the Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, Streptococcus agalactiae and Streptococcus dysgalactiae were 160 mg · L-1,80 mg · L-1,1 280 mg · L-1,640 mg·L-1 and 640 mg · L-1 respectively;The minimal inhibitory concentrations( MIC) of hydroquinone for these five bacteria were 640 mg · L-1 ,80 u,g · Ml-1,more than 1 280 mg · L-1,80 mg · L-1 and 80 mg · L-1 respectively; The minimal inhibitory concentrations ( MIC) of methyl gallate were 12 800 mg · L-1, more than 1 280 mg · L-1 , 640 mg · L-1 ,more than 1 280 mg · L-1 and more than 1 280 mg · L-1 respectively. The analysis of the results of orthogonal experiment showed that the sequence of the antibacterial effects of pyrogallol, hydroquinone and gallic acid methyl ester on Staphylococcus aureus, Staphylococcus epidermidis,Escherichia coli and Streptococcus agalactiae were pyrogallol,hydroquinone and methyl gallate;the sequence of the effects on Streptocuccus dysgalactiae were hydroquinone, methyl gallate and pyrogallol. The variance analysis showed that there existed significant differences between the factor levels of the three components on Staphylococcus aureus, Staphylococcus epidermidis and Escherichia coli(P<0. 05) while there existed no significant differences on Streptococcus agalactiae and Streptococcus dysgalactiae. Conclusion: Hydroquinone has good in vitro inhibitory effect on the main pathogens of cow mastitis and the best prescription of the three components is A3B3C2.关键词
