Abstract
Objective To observe the effects of triehostatin A (TSA) on the expression of p53 and p21 in thyroid squamous cell carcinoma cell line SW579 and lo explore its mechanism. Methods SW579 cells were cultured in vitro and divided into control, 50, 100, 200 and 400 nmol/L TSA groups randomly. The anti-proliferative effect of TSA against SW579 cells was detected by MTT. The p53 and p21 mRNA expression levels were determined by RT-PCR and p53 and p21 protein expression levels were detected by Western blot. Results The proliferation index decreased markedly with a dose-dependent manner. Compared with control groups, the expression levels of p21 mRNA and proteins of p21 and p53 were significantly elevated in various TSA groups, while p53 mRNA level was unchanged. Conclusion TSA can inhibit the proliferation of SW579 in a dose-dependent manner and increase p53 and p21 protein levels that may be the inhibit-pro-liferarion and induce-differentiation mechanism of TSA on thyroid squamous cell carcinoma cell line in vitro.关键词
