Influence of traditional Chinese medicine prescription for reinfocing Pi Qi on Th1/Th2 imbalance in rat asthma model and underlying mechanism

Abstract
Objective To investigate the effect of traditional Chinese medicine (TCM) prescription for reinfocing Pi Qi on regulation Thl/Th2 imbalance and its mechanism in the rat asthmatic modek. Methods SPF male Wistar rats were randomly divided into normal control group, asthmatic group, normal saline group, therapeutic group of Dexamethasone, treatment group of TCM prescription for reinforcing Pi Oi. Immune adjustment influence of medicine from reinforcing the spleen on Thl/Th2 imbalance was observed. The number of total cells, percentages of eoainophil (EOS) , lymphocyte, neutrophil were determined in bronckoalveolar Lavage fluid (BALF). Content of IL-4 and IFN-γ were detected by ELASA assay. The GATA-3 and T-bet expression in lung were detected by Western blot assay. Results Compared with normal group, the number of total cells, percentages of EOS, lymphocyte, neutrophil and level of IL-4 were all significantly increased in the BALF from rat of model group and normal saline treatment group, and the level of IFN-γ was dramatically decreased (P<0.01). However, the number of total cells, percentages of EOS,lymphocyte, neutrophil and level of IL-4 were markedly decreased, and level of IFN-7 was dramatically increased after administration of TCM prescription for reinforcing Pi Qi and Dexamethasone ( P < 0.01). The expression of GATA-3 was down-regulated, and the expression of T-bet was up-regulated. Conclusion There is immune modification effect of TCM prescription for reinforcing Pi Qi on Thl/Th2 imbalance, which may be acted by regulating balance expression of GATA-3 and T-bet.关键词
