鸭肝炎病毒Ⅰ型基因片段的原核表达及单克隆抗体的制备

Abstract
A pair of primers were designed according to the Genebank-reported complete genome of duck hepatitis virus I (DHV-1) ,and a specific gene segment of DHV-1 was obtained by RT-PCR. The segment was cloned into the expression vector pET-28a,then the recombinant plasmids were transfected into Rosetta-gami(DE3) .induced by IPTG and purified by the Ni-NTA,and thus the recombinant fusion protein(20 kD or so)was obtained. The BALB/c mice were immunized with this recombinant protein,and a monoclonal antibody cell strain of DHV-1 was produced. The Western-blot analysis and ELISA showed that the recombinant protein of DHV-1 could react with DHV-1 positive serum and DHV-1 monoclonal antibody. Also the DHV- I monoclonal antibody could detect natural protein of DHV-1 through western-blot assay.关键词
