青稞高分子量麦谷蛋白亚基SDS-PAGE分析
Analysis of HMW-GS in Hulless Barley by SDS-PAGE

Abstract
87 hulless barley varieties (strains) and 13 wild barley accessions were analyzed by SDS-PACE. The result showed that the HMW-GS bands were simple and lack of variance, with most materials had just one band and few materials had two bands; only 3 band positions in total occurred almost between 7 subunit and 12 subunit of Chinese Spring, a reference wheat cultivar. Thus the temporary names were given according to their respective migration distance in gel by comparing with the bands of the wheat cultivar Chinese Spring (CS): the band slightly lower than the 7 subunit of CS was named as A band; the band slightly lower than A band and much higher than 8 subunit of CS was named as B band; the band between 8 and 12 subunit of CS was named as C band. A, B and C band corresponded to A, B and C subunit, respectively. Four band patterns, A, B, C and AC were detected in 87 hull-less barley materials with each pattern containing 56, 1, 27 and 3 accessions and taking the ratio of 64. 4% , 1. 1% , 31. 0% and 3.4% , correspondingly. A, B and C pattern were single band patterns,and AC pattern was double band pattern; the AC pattern occurred when A pattern seeds and C pattern seeds existed in one material. These data indicates that the HMW-GS in hulless barley and wild barley may have single composing, lacking variation and lower polymorphism. The polymorphism study of high molecular weight glutenin subunits ( HMW-GS) in hulless barley can provide the basis for the flour milling and quality improving.关键词
