原子力显微镜观察青蒿琥酯对人胃癌细胞株SGC-7901膜表面形貌的影响
Effects of artesunate on membrane surface morphology of human gastric cancer SGC-7901 cells observed under atomic force microscope
摘要
Abstract
AIM; To delel the membrane surface morphology of cancer cells lrealed wilh arlesunale (ART) under alomic force microscope ( AFM). METHODS; Human gaslric cancer SGC -7901 cells were cullured and lrealed wilh different concenlralions of ART for 24 h. The membrane surface morphology, three - dimensional slruclures and ullra-slruclural changes of SGC -7901 cells were observed under AFM. The apoplosis of SGC -7901 cells was delecled by flow cylomelry. RESULTS; The AFM images revealed lhal the cell nuclear area was full and the surface of cell membrane was flal and smooth in conlrol group. Compared wilh conlrol group, the cell nuclear area collapsed and suffered alrophy, and the membrane surface had more pores in ART lrealmenl groups. More pores were observed and the diameler of the pores was increased wilh the increase in ART concenlralion. The cell membrane ullraslruclure showed lhal the panicles in conlrol group had an intensive distribution, some were cord - shaped and some gathered into a mass. The panicles in ART treatment groups were fewer and distributed sparsely. Moreover, there were obvious lacunae in the surface of cell membrane. Quantilive measurement found lhal the height of cell nucleus area was decreased, and the surface root mean square roughness (Rq) and the surface average roughness (Ra) became smaller in ART trealment groups compared wilh control group. The apoplolic rales of the cells in ART lrealmenl groups were increased wilh the increase in ART concenlralion, and were all significantly higher than lhal in conlrol group. CONCLUSION; Our findings emphasize the significance of AFM in exploring the changes of the membrane surface morphology al nano - scale resolution following the lrealmenl wilh anticancerdrugs, which could nol only idenlify the specific characlerislics of morphological changes of the Lumor cells, bul also provide micromorphological references Lo reveal the mechanisms of anlicancer drugs.关键词
