抗坏血酸对癫痫大鼠海马分子伴侣介导的自噬激活的影响
Effect of ascorbic acid on chaperone-mediated autophagy in hippocampus of rats with seizures

Abstract
AIM; To invesligale the role of chaperone - medialed aulophagy (CMA) and oxidalive slress in hippocampal injury induced by seizures and the neuroproleclive effecl of anlioxidanl ascorbic acid ( AA) on epileplic rals. METHODS; The rals were randomly divided inlo conlrol group, group with 24 h seizures induced by pilocarpine, group with 24 h seizures and AA prelrealmenl, and group of AA Irealmenl alone. Bolh mRNA and prolein levels of lysosome - as-socialed membrane prolein Lype 2a ( LAMP2a) were lesled by RT - PCR and Weslern blolling Lo evaluate the aclivily of CMA. The levels of malondialdehyde (MDA) and superoxide dismulase (SOD) were also measured by chemical melhods. RESULTS: The significant increases in mRNA and prolein levels of LAMP2a were found in hippocampus of the rals with 24 h seizures. The level of MDA highly increased and the level of SOD obviously decreased in the rals with seizures. AA prelrealmenl significantly decreased the levels of LAMP2a and MDA induced by seizures, but increased the aclivily of SOD. CONCLUSION: Bolh CMA and oxidalive slress are aclivaled in hippocampal injury induced by seizures. AA inhib-ils CMA and reduces oxidalive slress aclivaled by seizures by reducing LAMP2a and MDA, and has a polenlial anliepileplic function.关键词
