Abstract

The genetic evolution analysis was conducted for the glycoprotein genes in two isolated strains designated as 20101008 and 20101032 respectively of infectious hematopoietic necrosis virus(IHNV)in order to reveal the phylogenetic relationships of our strains and other strains in different countries and regions.The glycoprotein genes were amplified by RT-PCR and sequenced after being cloned directly into the pMD18-T.The sequences of G genes of 20101008 and 20101032 were compared with the sequences of other IHNV strains in GenBank by DNAStar and MEGA4.0 software.The results showed that the nucleotide and the deduced amino acid identities on G gene of our two strains were 98% and 81.9% respectively.Further analysis of the deduced amino acid sequences indicated 35 and 20 of amino acid residues of G gene were substituted in 20101008 and 20101032.They shared closer identity with strains isolated from Japan and South Korea,and the rate of nucleotide and amino acid homology of G gene were 93.3%~96.4% and 75.9%~84.6% respectively.The two strains 20101008 and 20101032 had the closest genetic relatives and belonged to the same branch and they belonged to genotype U.However,the G gene diverges from known strains at either nucleotide level or amino acid level.
