地西他滨联合丙戊酸钠对U937细胞hPer3基因表达调控的影响
Synergistic effect of decitabine and valproic acid on hPer3 gene expression in leukemia U937 cells

Abstract
Objective To investigate the synergistic effect of decitabine (DCA) and valproic acid (VPA) on human period3 (hPer3) gene expression in leukemia U937 cells. Methods The groups were set as follows ; group A ( control group) ; groups B and C ( DCA 1, 4 pjnol ? L"1) ; group D(VPA 2 mmol ? L'1) ; groups E and F (DCA 1, 4 jjimol ? L"1 + VPA 2,2 mmol ? L'1) . The cells were treated by drug for 48 h. Then the inhibitions rates, apoptosis rates, promoter methylation status of hPer3 gene,hPer3 mRNA were detected by MTT, Annexin V/PI, MS -PCR and qRT -PCR. Results hPer3 mRNA, inhibitions rates, apoptosis rates,CD117 MFI.CD14 expression in groups E and F were significantly higher (or lower) than their corresponding concentration single drug groups (/* < 0.01). Conclusion There was an enhanced antileu-kemia activity of combinations of DCA and VPA. The combination of DCA and VPA could induce the expression of hPer3 gene.关键词
