Abstract
AIM To establish a method for determining the concentration of doxofylline in human serum by ultra high performance liquid chromatography (UPLC). METHODS The column was Waters Acquity C18(50 mm× 2.1 mm, 1.7 μm), the mobile phase was acetonitrile-water in gradient programs at flow rate of 0.2 mL· min-1, and the caffeinewas used as internal standard. The temperature of column was set at room temperature and the detection wavelength was set at 273 nm. RESULTS The calibration curves were linear in the range of 0.18-23.20 mg·L-1 for doxofylline;the linear regression equation was y= 0.181 8x + 0.005 l(r = 0.999 9, n = 10);the RSD of intra-day and inter-day was below 15 %; the recovery rates were 94.70% - 106.16%. CONCLUSION The method is sensitive, rapid and accurate, which is usable for monitoring of doxofylline and pharmacokinetic studies.关键词
