摘要

Abstract

Objective:To deled the inhibilive effecl on multiple myeloma (MM) cancer stem cells(CSCs) by ABCG2 monoclonal anlibody (McAb) in combination wilh Fe3O4 nanoparticles-paclitaxel (NPs-PTX) in vitro. Methods; MM CSCs (CD138~ CD34 ~ ) were sorled from human MM cell lines NCI-H929 and RPMI8226 through the immune magnetic beads sorling method. Growlh inhibitive effect of ABCG2 McAb in combination with NPs-PTX on MM CSCs was delecled by CCK-8 assay. The migration rale of MM CSCs was delecled by transwell assay. Effecls of cell cycles and apoplosis were also delecled by flow cylomelry (FCM). Results;The proliferation of NCI-H929 and RPMI8226 cells were suppressed after trealmenl wilh ABCG2 McAb in combination wilh NPs-PTX for 24 hours. The growlh inhibilion rale was 70% in lhe NCI-H929 cells or 80% in lhe RPMI8226 cells. There was slalislical significant difference (P<0.05) between lhe combined utilization of ABCG2 McAb and NPs-PTX group (80% or 70% ) in contrast to lhe utilization of NPs-PTX group alone (55% ). As to lhe rate of migralion, ABCG2 McAb in combination wilh NPs-PTX group was 0. 6% but that of NPs-PTX group was 1.7%. The difference was statistically significant (P < 0. 01). ABCG2 McAb in combination with NPs-PTX could effectively induce MM CSCs apoplosis, and early apoplosis rate reached 43. 15%. Compared to NPs-PTX group (18. 35% ), there was slalislical significant difference (P <0. 05) in the combined group. In addilion, ABCG2 McAb togelher with NPs-PTX could make MM CSCs cell cycle blocked in G/M period. Conclusion: ABCG2 McAb in combination with NPs-PTX has lhe function of inhibiting MM CSCs in vitro, which may associate with induction of cell apoptosis and interruption of cell cycle.
