HIF-1α、MT、Slug和E-cadherin在人乳腺癌中的表达及其意义
The expressions and significances of HIF-1α, MT, Slug and E-cadherin in human breast cancer

Abstract
Objective:To investigate the expressions and significances of HIF-1 α, MT, Slug and E-cadherin in human breast cancer. Methods: Immunohistochemistry was used to analyze the expression of HIF-1 α, MT, Slug and E-cadherin in 69 tissue samples of breast cancer patients without any radiotherapy, chemotherapy and hormone therapy. The mRNA expressions of HIF-1 α., MT, Slug and E-cadherin in breast cancer cell lines MCF-7 and MDA-MB-231 were detected by RT-PCR analysis. Results: In 69 tissue samples of breast cancer, the positive expression rates of HIF-lα, MT, Slug and E-cadherin were 71. 01% , 42. 03% , 43. 48% and 50. 72%. The expressions of HIF-1 α, Slug and E-cadherin have significant correlations with TNM stage and axillary lymph node metastasis ( P <0. 05 ), the expression of MT has a significant correlation with TNM stage ( P < 0. 05 ). In 69 tissue samples of breast cancer, the expression of HIF-1 α has a positive correlation with that of MT ( r =0. 285,P <0. 05 ), the expressions of MT and Slug were significantly positively correlated ( r =0. 379, P <0. 01 ), and the expression of Slug was significantly negatively correlated with that of E-cadherin ( r=- 0. 422,P <0.01 ) in breast cancer. RT-PCR analysis showed that HIF-lct, MT, Slug and E-cadherin mRNA were expressed in breast cancer cell lines MCF-7 and MDA-MB-231. The mRNA expression levels of HIF-lα, MT and Slug was higher and the expression levels of E-cadherin was lower in MDA-MB-231 cell line than in MCF-7 cell line( P<0.05 ). Conclusion:The expressions of HIF-1 α., MT, Slug and E-cadherin might be used as markers for development of breast cancer.关键词
