Abstract

To study the effects of ivermectin on survival rate and serum cytokine secretions in lipopolysaccharide(LPS)-induced endotoxemic mice,C57BL/6 mice were given intraperitoneally with LPS at a lethal dose of 32 mg/kg,and ivermectin of different doses(1,2 and 4 mg/kg) were pre-treated with an orallyadministration 2 h before LPS challenge. Survival of mice was monitored for 7 days twice a day following administration of LPS. The levels of turnout necrosis factor-α(TNF-α),interleukin-lβ(IL-1β) and interleukin-6(IL-6) in serum of the mice were assessed by sandwich ELISA method,and nitric oxide(NO) levels were measured by nitric acid reduction method at different time following LPS administration. The results showed that ivermectin significantly increased the survival rate in LPS challenged mice, decreased serum levels of TNF-α,IL-1β, IL-6 and NO after LPS challenge. These observations indicated that ivermectin can improve survival rate of endotoxemia mice induced by LPS,which may be mediated by the inhibition of inflammatory cytokine and mediator production.
