Expression of recombinant bovine interferon-a with antiviral activity by using intein induced expression system
摘要
Abstract
To achieve high-level expression of soluble recombinant bovine interferon-a(BoIFN-a) in Escherichia coli ,the Ssp DnaB intein(SDI) coding sequence was merged into the upstream of the BoIFN-a cDNA,and then cloned into the expression vector pMAL-c2X to construct the intein expression plasmid pMAL-SDI-BoIFN-a. The recombinant expression plasmids were transformed into E. coli Rosetta and induced by IPTG. The result showed that the fusion protein was expressed in the form of solubility and more pure proteins can be obtained by Amylose Resin column purification. The purified protein shaved the antiviral activity in MDBK-BVDV(bovine viral diarrhea virus) system.关键词
