摘要

Abstract

Objective To examine morphological changes of peripheral blood mononuclear cells (PBMCs) due to inflammation by atomic force microscopy (AFM). Methods A total of 66 uremic patients on haemodialysis (MHD group),30 uremic patients without initiation of haemodialysis who had not experienced cardiovascular events (WHD group), and 20 healthy volunteers were selected as study subjects. A-mong the 66 MHD patients,36 patients had previous cardiovascular events (MHD1 group) and 30 patients didn't (MHD2 group). PBMCs were separated by using lymphocyte separation medium,and observed under an inverted light microscope. AFM was used to analyze cell volume, mean height,half-maximum amplitude,average roughness,and the diameters of surface granules on PBMCs. Levels of TNF-a and IL-10 in cell supematants were detected by ELISA. Results No significant difference was observed in PBMC morphology between groups by light microscopy. AFM images revealed that PBMCs in uremic patients had granular processes or caveolae of uneven size distributed over the cell surfaces,and were larger than the cells from healthy volunteers in volume,mean height,half-maximum amplitude,and average roughness. Moreover,uremic patients had higher TNF-a levels but lower IL-10 levels comparing with healthy individuals (P< 0.05). These differences between the WHD group and MHD group, MHD1 group and MHD2 group (P < 0.05) were also significantly. Cell volume,mean height, half-maximum amplitude,and average roughness were positively correlated with TNF-a levels but negatively correlated with IL-10 levels in uremic patients on haemodialysis. Conclusion PBMCs morphology was associated with inflammatory state in uremic patients on haemodialysis.
