甲基莲心碱对2型糖尿病模型大鼠作用的代谢组学研究
Effects of neferine on type 2 diabetic mellitus rats by metabonomics method

Abstract
Aim To study whether neferine influences urinary metabolites of type 2 diabetic mellitus rats. Methods The rats were fed with high-sucrose and high-fat chow for 4 weeks, then were injected intraper-itoneal(ip) with STZ (30 mg ? kg-1). One week later, rats with fasting blood sugar levels higher than 7. 8 mmol ? L-1 were defined as type 2 diabetes mellitus (T2DM) rats. Neferine treated T2DM rats were injected intraperitoneal (ip) with neferine (20 mg ? kg-1? d-1) for 4 weeks. Fasting blood sugar of all rats was measured after 4 weeks. 24 h urine samples from rats were collected and prepared. All the separations were performed on UPLC/QTOF-MS system. Results The blood glucose values in neferine treated T2DM ratswere decreased in comparison with those in T2DM rats (P < 0. 05). Dihydroxyfumaric acid in urinary metabolite profiles of neferine treated T2DM rats was decreased compared with that in T2DM group. Increase of PGI2, LXA4 and LXB4 was revealed in the urinary metabolite profiles of neferine treated T2DM group than that in T2DM group. Conclusions Neferine can decrease blood glucose and change urinary metabolite profiles of T2DM rats. The mechanism of neferine may be associated with antioxidant stress and anti-inflammatory effect.关键词
