Abstract

Aim To investigate effects of hydrogen-rich saline on hippocampus mitochondrial permeability transition pore (mPTP) and apoptosis of rats with global cerebral ischemia-reperfusion (I/R) injury. Methods Ninety-six healthy male SD rats(250 ~ 300 g) were randomly divided into six groups (n = 16 each): sham operation group (S), cerebral ischemia-reperfusion group (IR), normal saline group (NS), hydrogen-rich saline group (H), atractyloside group (A) and hydrogen-rich saline + atractyloside group (HA). Global cerebral I/R was produced by four-vessel occlusion method. Bilateral vertebral arteries were cauter-ized. Bilateral common carotid arteries were occluded for 15 min and followed by reperfusion. In group H and HA, hydrogen-rich saline 5 ml · kg-1 was intraperito-neally injected immediately after reperfusion, while the equal volume of normal saline was injected in the other four groups. The animals in group A and HA were given atractyloside by intracerebroventricular injection (ICV) at 10 min before reperfusion, while rats in group NS and H were injected with normal saline by ICV. The animals were sacrificed at the end of 24 h reperfusion. The mPTP opening was assayed by spectropho-tometry . Cytochrome C (Cyt C) protein expression in the mitochondria and cytoplasm was determined by western blot. Bcl-2 and Bax protein expression was determined by immuno-histochemistry. The hippocampus mitochondrial ultrastructure was examined with transmission electron microscope. Results Compared with group S, mPTP opening was enhanced, Bcl-2 and Bax expression was significantly higher, mitochondrial Cyt C protein expression was lower, cytosolic Cyt C was higher in the other groups (P < 0. 05). mPTP opening was decreased, Bcl-2 expression was higher, Bax expression was lower, mitochondrial Cyt C protein expression was lower, cytosolic Cyt C was higher in group H than in group IR(P < 0. 05). Compared with group H, mPTP opening was increased, Bcl-2 expression waslower, Bax expression was higher,mitochondrial Cyt C protein expression was lower, cytosolic Cyt C was higher in group HA(P < 0. 05). Microscopic examination showed less damage in group H than in group IR. The protection provided by hydrogen-rich saline was abolished by atractyloside. Conclusion Hydrogen-rich saline attenuates global cerebral I/R induced injury, decreases Cyt C protein releasing and suppresses mitochondrial apoptotic signal transduction by inhibiting mPTP opening.
