Effect of Baixiangdan on the expression of 5-HTR2C and the concentration of IP3 in neonate rat hippocampal neurons cultured in vitro

Abstract

Aim To explore the intervention mechanisms of baixiangdan capsule by observing the changes of the expression level of 5-HTR2C and the concentration of IP3 in neonate rat hippocampal neurons culturedin vitro. Methods Premenstrual syndrome ( PMS ) model rats with liver-qi invasion were induced by multiple emotional stimulations and baixiangdan capsule was administered after PMS induction. The medicatedserum was used to neonate rat hippocampal neurons . The expression level of 5-HTR2C in the neurons was e- valuated by Western blot and the concentration of IP 3 was measured with the Elisa method. Results Compared with control group , both the protein expression of 5-HTR2C and the concentration of IP 3 of PMS liver-qi invation group were significantly higher in neurons (P < 0. 01, P < 0. 01 ). After treated with baixiangdan medicated serum, the expression of 5-HTR2C and the concentration of IP3 in hippocampal neurons were significantly improved(P<0. 01,P<0. 01). ConclusionsPMS model rats with liver-qi invasion serum can up-regulate the expression of 5-HTR2C in hippocampal neurons and increase the concentration of the second messenger IP3. Baixiangdan capsule might partially play its role in ameliorating PMS by regulating the expression of 5-HTR2C and changing the concentration of IP3.
