Abstract
Objective To establish a sensitive and rapid high performance liquid chromatographic - tandem mass spectrometric (HPLC -MS/MS) method for the simultaneous determination of amantadine hydrochloride and chlorpheniramine maleate in human plasma. Methods The C18 column was used with water (containing 0.5‰ formic acid): acetonitrile: methanol(18: 32: 50) as the mobile phase and finasteride as the internal standard. The concentration of amantadine hydrochloride and chlorpheniramine maleate in human plasma was determined by HPLC - MS / MS with selected reaction monitoring (SRM) mode via electrospray ionization (ESI) . Results The concentration of amantadine hydrochloride and chlorpheniramine maleate were linear in the range of 3.9-375.0 ng/mL and 0.2-19.2 ng/mL,respectively. The relative recovery rates of amantadine hydrochloride were 106.4% (low), 94. 9% (middle) and 93.1% (high), respectively and its intra and inter - day RSD were 2. 6% ,3. 2% ,4. 1% and 4. 3% ,5. 4% ,5. 6% respectively. Its lowest limit of quantification was 3.9 ng/mL with RSD 3.1%. The relative recovery rates of chlorpheniramine maleate were 98. 9% (low), 97.9% (middle) and 101.8% (high) respectively and its intra and inter-day RSD were 8. 5% , 8. 0% ,4. 1% and 6. 9%, 5. 5%, 5.6% ,respectively. Its lowest limit of quantification was 0.2 ng/mL with RSD 1.7%. Conclusion The established method is highly sensitive, simple and accurate, which can be suitable in simultaneous quantification of amantadine hydrochloride and chlorpheniramine maleate in human plasma and may be used for the study of pharmacokinetics of amantadine hydrochloride and chlorpheniramine maleate.关键词
