苦参碱对喉鳞癌细胞株Hep-2侵袭转移的影响

Abstract
Objective To study the inhibitory effects of Matrine on invasion and metastasis of laryngeal squamous cell carcinomas cells Hep-2. Methods Hep-2 cells were cultured in vitro and treated by 0 g/L, 0.1 g/L, 0.2 g/L and 0.3 g/L Matrine. Then cell adhesion assay, cell migration assay and cell invasion assay were used to observe the effects of Matrine on adhesion, migration and invasion of Hep-2 cells, and RT-PCR were performed to evaluate the MMP-2 and MMP-9 mRNA expression levels. Results Contrasting to control group, 0.1 g/L, 0.2 g/L and 0.3 g/L Matrine not only could effectively inhibit the adhesion, migration and invasion of Hep-2 cells(p< 0.01), but also could down- regulate the expression of MMP-2 and MMP-9 mRNA(p < 0.01) Conclusion By down-regulating the expressions of MMP-2 and MMP-9 mRNA, Matrine can inhibit the capability of adhesion, migration and invasion of Hep-2 cells.关键词
