人结肠癌动物皮下实体瘤模型的建立
Establishment of Subcutaneous Solid Tumor Mice Models of Human Colon Carcinoma

Abstract
[Objective] To establish subcutaneous solid tumor mice models of human colon cancer which have clear clinical backgrounds. [Methods] 12 clinical colon cancer specimens were first transplanted to the subcutaneous tissue of right shoulder in NOD-Scid mice by tissue transplantation, Then the passage 2 of the subcutaneous tumor in each cases were implanted contiguously into NOD-Scid mice and BALB/C nude mice, respectively, which were subcultured to passage 5 in mice. The time and rate of tumor formation, HE staining and immunohistochemical examination of CDX-2 and CK20 were observed during the experiment. [Results] The rates of subcutaneous tumor formation of passage 3 and 5 were 92% and 58% in NOD-Scid mice, while 75% and 50% in nude mice, respectively. The pathological results showed that cells in tumor tissue were gland-like and arranged irregularly, in which pathological nuclear fission was observed. The immunohistochemical stain of CDX-2 and CK20 in cells were positive. [Conclusions] The subcutaneous solid tumor models of human colon cancer in NOD-Scid and nude mice were established preliminary, providing a technological foundation for further exploring the pathogenesis and therapy strategy of colon cancer.关键词
