槲皮素对人肾癌细胞株细胞增殖、侵袭及其VEGF表达的影响
Effects of quercetin on proliferation, invasion and VEGF protein expression on human renal cell cancer of ACHN cell line
摘要
Abstract
Objective To study the effect of quercetin on the proliferation and invasion on human renal cell cancer of ACHN cell line, and investigate the probable molecular mechanisms. Methods Different concentrations of quercetin were used to treat ACHN cells cultured in vitro, the inhibition of proliferation of ACHN cell was studied by methyl thiazolyl tetrazolium ( MTT ) assay. The abilities of cell invasion was evaluated by Transwell chambers, and the expression of vascular endothelial growth factor ( VEGF ) was detected by Western blot analysis. Results After quercetin treatment, the proliferation and invasive ability of ACHN cells were obviously decreased, which showed a dose-and time-dependent manner( P <0. 05 ). Meanwhile, quercetin could down-regulate the expression of VEGF protein in a dose-dependent manner. Conclusion Quercetin can inhibit the proliferation and invasion of ACHN cells by down-regulating VEGF expression.关键词
