熊果酸抑制脂肪酸合酶及诱导HepG2细胞凋亡机制研究
URSOLIC ACID INDUCED HEPG2 CELLS APOPTOSIS VIA INHIBITING FATTY ACID SYNTHASE
摘要
Abstract
The widely present plant chemical ursolic acid(UA) has important anti-cancer activity.UA is a natural inhibitor of fatty acid synthase(FAS) which is highly expressed in cancer cells.It was found in MTT assay that UA inhibited cell growth with an IC50 of 68.23 μmol·L-1.Hoechst 33258 fluorescence microscopy revealed that UA induced apoptosis in HepG2 cells dose-dependently.UA addition reduced significantly FAS expression.After sodium palmitate addition to cells incubated with UA,cellular viability was restored.It is concluded that tumor suppression activity of UA is due to its inhibitory activity against tumour cell FAS,leading to blockade of fatty acid synthesis.The lack of fatty acids for cellular metabolism triggers cancer cell apoptosis.关键词
