Abstract
Objective To study the activated platelet delivers the sphingosine 1 phosphate) which has some influence on platelet's function during conservation. Methods Using enzyme linked immunosorbent assay to observe the create regularity of S1P. Activation and apoptosis of platelet were detected by flow cytometry. Results As to conservation time, the content of S1P continued to increase. The contents of SIP (μmol/Dwere 1. 38±0.17,2. 07±0. 25,2. 78±0.31,3. 53 ± 0. 28 and 4. 56 ± 0. 46 from 1 d to 5 d preservation respectively. We observed the changes of platelet's function after adding exogenous S1P with different concentration too. The results displayed that the ratio of activation and apoptosis increased with SlP concentration, and had disparity in different groups. Platelets' structure changed too. Conclution As extended to save time, S1P level in platelets' plasma gradually increase, and platelets' function gradually reduce. The results suggest that phospholipid metabolism with biological activity released by platelet may exert a certain influence on platelet function, which could be an important reason to cause platelet transfusion refractoriness.关键词
